5-Hydroxytryptophan (5-HTP) is an intermediate metabolite in the biochemical synthesis of serotonin from the essential amino acid tryptophan. The conversion of tryptophan to 5-HTP involves the enzyme tryptophan hydroxylase (TPH), which exists in two isoforms: TPH1 and TPH2. TPH1 is mainly expressed in the peripheral tissues, while TPH2 is predominantly found in the central nervous system. Once synthesized, 5-HTP is further decarboxylated by aromatic L-amino acid decarboxylase (AADC) to produce serotonin (5-hydroxytryptamine, 5-HT). This biogenic amine plays a crucial role in regulating mood, cognition, and various physiological processes. Serotonin can also be acetylated and methylated to produce melatonin, a hormone involved in regulating sleep-wake cycles. Given its central role in serotonin biosynthesis, 5-HTP is often used in the treatment of conditions like depression, anxiety, and insomnia. Recent biotechnological advances have focused on optimizing the production of 5-HTP through engineered enzyme systems to meet increasing demand.
