ARL8A and ARL8B are members of the ADP-ribosylation factor-like (ARL) family of small GTPases, which play important roles in intracellular trafficking and vesicular transport. The structure of ARL8A and ARL8B consists of a conserved GTPase domain that binds to guanine nucleotides (GDP and GTP) and a C-terminal lipid modification site that allows their association with cellular membranes. They share a high degree of sequence similarity, with ARL8B being the closest homolog of ARL8A. Functionally, ARL8A and ARL8B are involved in the regulation of lysosomal positioning and trafficking. They are primarily localized to late endosomes and lysosomes, where they interact with various effector proteins to control the movement of these organelles along microtubules. ARL8A and ARL8B have been implicated in mediating lysosomal fusion events, lysosomal tubulation, and lysosome-related processes such as autophagy.
