          CD26 Antibody (BMS143) in Flow

          Knockout of CD26 (DPP4) was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR49285_LV) and LentiArray Cas9 Lentivirus (Product # A32064).           Product Specific Information

          Description: CD26 is a 110 kD type II membrane protein encoded by the DPP4 gene. It is an antigenic enzyme expressed on the surface of most cell types and is associated with immune regulation, signal transduction and apoptosis. It also plays an integral role as a costimulatory molecule in T cell activation. CD26 may interact with extracellular matrix proteins such as fibronectin or collagen, CD45 and ADA.

          Immunohistochemistry:

          The antibody can be used to stain cryostat sections. Recommended for BMS143 is the alkaline phosphatase-anti-alkaline-phosphatase (APAAP), or peroxidase anti-peroxidase (PAP) procedures or the three stage immunoperoxidase technique on acetone-fixed cryostat sections.

          Flow cytometry:

          BMS143 is also suitable as primary antibody in staining for FACS analysis, BMS143FI can be used for direct staining of cells, BMS143BT can be used in a two-step procedure using the biotin-(strept-)avidin system. Recommended dilution is 1:20 as determined on A704 cells.

          Immunoprecipitation:

          BMS143 is also suitable for immunoprecipitation. Optimal dilutions should be determined by the individual laboratory for each application.

          Target Information

          CD26 (dipeptidyl peptidase IV, DPP IV), adenosine deaminase (ADA) binding protein) is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family. CD26 is expressed on lymphocyte cells and is upregulated during T-cell activation. CD26 is also expressed on activated B cells and natural killer cells and abundantly on epithelia. CD26 is implicated in a variety of biological functions including T-cell activation, cell adhesion with extracellular matrix such as fibronectin or collagens, and in HIV infection. CD26 identical to adenosine deaminase complexing protein-2, and to the T-cell activation antigen CD26. Further, CD26 is an intrinsic membrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. Alterations in CD26 peptidase activity are characteristic of malignant transformation, and the enzymatic activity increases dramatically with tumor grade and severity. CD26 is expressed in various blood cell types, but also in cells that are histogenetically related to activated fibroblasts. Alterations in CD26 density have been reported on circulating monocytes and CD4+ T cells during rheumatoid arthritis and systemic lupus erythematosus.
