Normal human peripheral blood cells were unstained (left) or stained with CD305 (LAIR1) Monoclonal Antibody, NovaFluor Blue 660-120S (right). All cells were co-stained with CD56 (NCAM) Monoclonal Antibody, eFluor 450 (Product # 48-0566-42). Total viable cells in the lymphocyte gate were used for analysis, as determined by LIVE/DEAD Blue (Product # L34962). Data was acquired on a 5-laser Cytek Aurora and unmixed with autofluorescence extraction.

Description: This NKTA255 monoclonal antibody reacts with human CD305, which is also known as leukocyte-associated Ig-like receptor (LAIR) 1. CD305 is a type I transmembrane glycoprotein that possesses an extracellular Ig-like domain and two intracellular ITIMs. CD305 is expressed on nearly all immune cells, including T, NK, and B cells, as well as monocytes, dendritic cells, eosinophils, basophils, and mast cells. In humans, this molecule is also expressed on CD34+ hematopoietic progenitor cells and thymocytes. Several isoforms of CD305 exist due to alternative splicing, and their expression may differ between cell types. As an inhibitory receptor, CD305 blocks cytokine-mediated signaling, cell proliferation, and apoptosis. Studies have also identified collagens as the ligands for CD305.

Applications Reported: This NKTA255 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This NKTA255 antibody has been pre-titrated and tested by flow cytometric analysis of human peripheral blood cells. This can be used at 5 µL (0.8 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer .
NovaFluor dyes are not compatible with DNA intercalating viability dyes. Do not use viability dyes such as propidium iodide, 7-actinomycin D (7-AAD) and DAPI. Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes.
This NovaFluor conjugate has been updated to ship with CellBlox Plus Blocking Buffer (Cat. No. (C001T06F01)). This buffer contains formulation improvements over CellBlox. CellBlox Plus Blocking Buffer is required for optimal staining with NovaFluor conjugates and should be used in all experiments where NovaFluor conjugates are used. Whenever possible, we recommend adding CellBlox Plus Blocking Buffer to antibody cocktails/master mixes prior to combining with cells. Add 5 µL per sample (regardless of the number of NovaFluors in your panel) to use the antibody cocktail as intended. For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL of cell sample containing 10^3 to 10^8 cells.

Target Information
LAIR1 (CD305) functions as an inhibitory receptor that plays a constitutive negative regulatory role on cytolytic function of natural killer (NK) cells, B-cells and T-cells. Activation by Tyr phosphorylation results in recruitment and activation of the phosphatases PTPN6 and PTPN11. It also reduces the increase of intracellular calcium evoked by B-cell receptor ligation. May also play its inhibitory role independently of SH2-containing phosphatases. Modulates cytokine production in CD4+ T-cells, down-regulating IL2 and IFNG production while inducing secretion of transforming growth factor beta. Down-regulates also IgG and IgE production in B-cells as well as IL8, IL10 and TNF secretion. Inhibits proliferation and induces apoptosis in myeloid leukemia cell lines as well as prevents nuclear translocation of NF-kappa-B p65 subunit/RELA and phosphorylation of I-kappa-B alpha/CHUK in these cells. Inhibits the differentiation of peripheral blood precursors towards dendritic cells.
