CD4 Antibody (MHCD0404) in Flow

Flow cytometry histogram showing human mononuclear cells stained with anti-CD4 complexed with Alexa Fluor® 700 Zenon® labeling reagent. An antibody complex was made by using the Zenon® Alexa Fluor® 700 Mouse IgG1 Labeling Kit (Product # Z-25011) and mouse anti human CD4. Human mononuclear cells were stained with the antibody complex and analyzed by flow cytometry using a 633 nm laser.

CD4 Antibody — Figure 7 Endocytosis was necessary in the process of DRibbles-inflammasome response. ( a ) CypHer5E-labeled DRibbles and lysotraker Green-labeled lysosome were collocated in THP-1 cells after 6 h incubation, and the nucleus were labeled with DAPI. ( b ) The endocytosis frequency of DRibbles by THP-1 cells was measured by FACS. DCs were pre-treated with ExoA (1 mu M), Cytochalasin-D (Cyto-D, 1 mu g/ml), and NH 4 Cl (20 mu M) for 30 min, then incubated with CypHer5E stained DRibbles (100 mu g/ml) overnight. ( c ) Primed THP-1 cells were pre-treated with cytochalasin-D, ExoA, and NH 4 Cl for 1 h, then stimulated with ATP/LPS (5 mM)/(10 ng/ml) and DRibbles (100 mu g/ml) overnight. The supernatant IL-1 beta was detected by ELISA. ( d ) The frequency of IFN- gamma + CD8 + , IFN- gamma + CD4 + T cells induced by pp65 DRibbles at indicated doses were measured by ICS, and ( e ) pp65 protein (10 mu g/ml) and pp65 DRibbles (10 mu g/ml). Results are shown as the means+S.D. and representative of two or more independent experiments. PBMC is from 3 donors {TM}

R-phycoerythrin (PE) is a stable and highly soluble phycobiliprotein which provides maximal absorbance and fluorescence without susceptibility to internal or external fluorescence quenching, thus providing an exceptional quantum yields and molar extinction coefficients.

This antibody may be used, in combination with other indicators, for the diagnosis or prognosis of immunodeficiency diseases, including hypogammaglobulinemia, severe combined immunodeficiency (SCID), and the acquired immunodeficiency disease (AIDS).

Target Information

The CD4 antigen is involved in the recognition of MHC class II molecules and is a co-receptor for HIV. CD4 is primarily expressed in a subset of T-lymphocytes, also referred to as T helper cells, but may also be expressed by other cells in the immune system, such as monocytes, macrophages, and dendritic cells. At the tissue level, CD4 expression may be detected in thymus, lymph nodes, tonsils, and spleen, and also in specific regions of the brain, gut, and other non-lymphoid tissues. CD4 functions to initiate or augment the early phase of T-cell activation through its association with the T-cell receptor complex and protein tyrosine kinase, Lck. It may also function as an important mediator of direct neuronal damage in infectious and immune-mediated diseases of the central nervous system. Multiple alternatively spliced transcripts have been identified in this gene [RefSeq, July 2017].
