Description: The MRa4-1 monoclonal antibody reacts with CD49d, which is also known as the integrin alpha 4 subunit. This 150-kDa cell surface protein is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils in complex with either integrin beta1 or integrin beta7. Outside of the immune system, CD49d is also expressed on mast cells, neural crest cells, retinal cells, and dorsal root ganglion neurons. Integrins are responsible for lymphocyte homing to Peyer's patches and mesenteric lymph nodes, as well as neural development and regeneration. Several ligands have been reported for the integrin a4b1 complex, including VCAM-1 (CD106) and fibronectin.
The MRa1-4 antibody has been reported to inhibit fibronectin binding and lymphocyte migration.
Applications Reported: This MRa4-1 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This MRa4-1 antibody has been tested by flow cytometric analysis of rat splenocytes. This can be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

The integrin alpha 4 (also known as CD49d and ITGA4) belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha and beta chains. Alpha4 chain associates with either beta1 or beta7 chain. It has been demonstrated that the putative ligand-binding sites of both integrin alpha4beta1 and alpha4beta7 is located on the alpha4 chain. These ligands included Madcam, VCAM, and fibronectin. Madcam is known as the principal ligand for integrin alpha4beta7. Recently it was also demonstrated that HIV-1 envelope can mimic Madcam by binding to and signaling though integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
