          CXCL10 Antibody (MA5-33342) in ICC/IF

          Immunofluorescence analysis of C-X-C motif chemokine 10 was performed using 70% confluent log phase HT-29 cells and HT-29 cells treated with 50ng/mL IFN for 14h followed by 1x Protein Transport Inhibitor (PTI) treatment for 4h. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature.           Product Specific Information

          Product is supplied at 1 mL (100 µL/mL). Application Notes: Flow Cytometry - For intracellular staining activated plasmacytoid dendritic cells were fixed with 2% paraformaldehyde and permeabilized with 0.5% saponin. All steps were performed in presence of 0.5 M EDTA to avoid cell aggregation. Western Blot - Tissue was homogenized in loading sample buffer and subjected to SDS-PAGE. The band size is ~9 kDa. IHC-F - Tissue sections were fixed 10 minutes in acetone and pretreated with avidin/biotin blocking kit. As positive control chronically inflamed liver was used and as negative control normal liver and isotype matched antibody.

          Positive control: Acitvated plasmacytoid dendritic cells; Negative control: Normal liver.

          Target Information

          IP-10 (interferon-gamma induced protein 10 kD; in mouse, also called cytokine responsive gene 2, Crg-2, or mob-1) is a highly inducible, primary response gene that belongs to the CXC chemokine superfamily. It was first cloned in 1985. The biological functions of IP-10 are still unclear. Like Mig, IP-10 has no activity on neutrophils. Its functions include stimulation of monocytes, natural killer and T-cell migration, regulation of T-cell and bone marrow progenitor maturation, modulation of adhesion molecule expression as well as inhibition of angiogenesis. Mig and IP-10 share the same receptor, CXCR3.
