Calmodulin 2 (CALM2) is one of the three human genes encoding calmodulin, a highly conserved calcium-binding messenger protein critical for mediating various cellular processes. Calmodulin proteins are composed of two pairs of EF-hand motifs located at both the N- and C-termini, connected by a flexible central linker. Each EF-hand motif binds calcium ions, with the C-terminus possessing two high-affinity Ca2+ binding sites, while the N-terminus contains two low-affinity sites. Upon binding calcium, calmodulin undergoes a conformational change that enables it to interact with and regulate numerous target proteins, including kinases, phosphatases, ion channels, and other enzymes. Specifically, calcium/calmodulin (CaM) signaling pathways are involved in critical cellular functions such as muscle contraction, neurotransmitter release, and cellular growth. The interaction of CALM2 with different targets is essential for translating transient changes in intracellular calcium levels into specific cellular responses, making it a pivotal player in calcium signaling.
