Localizes Dsc1 present insuprabasal layers of interfollicular epidermisspecific cell layers (e.g. in hair follicle root sheath)Hassall bodies (spheroidal formations of densely packed reticulum-derived cells in the thymus).Reacts with 111 kD (form "a") and 103 kD (form "b") polypeptides (desmo-collin 1 splice forms, both at pI 5.2) from human epidermal desmosomes.Reactivity on cultured cell lines: Positive on primary keratinocytes, however, restricted to local piles of differentiated cells. All simple epithelia are negative.
The desmosomal cadherin desmocollin DSC1 is expressed in upper epidermis where strong adhesion is required. DSC1 is a type I membrane protein (4) required for strong adhesion and barrier maintenance in epidermis and contributes to epidermal differentiation.The DSC1 gene comprises 17 exons spanning approximately 33 kb on 18q12.1.
