          Product Specific Information

          The monoclonal antibody 74.1 recognizes all known forms of the 74 kDa intermediate chain subunit of mammalian cytoplasmic dynein. Cytoplasmic dynein is a microtubule-stimulated ATPase responsible for movement of membrane-bound organelles in a directed manner toward the minus end of microtubules, commonly found at the centrosome. Dynein is responsible for the transport of lysosomes, endosomes, phagosomes, peroxisomes, mitochondria, and vesicles from the endoplasmic reticulum destined for the Golgi. During mitosis, dynein is responsible for positioning the spindle apparatus as well as being localized to the kinetochores to facilitate chromosome movement.The 74.1 antibody recognizes human, mouse, rat, bovine, sheep, Xenopus, porcine, and Drosophila dynein intermediate chain.

          This 74.1 antibody has been tested by immunocytochemistry of methanol-fixed and permeabilized cells and can be used at less than or equal to 1 µg/mL. This 74.1 antibody has been tested by western blot of reduced and non-reduced HeLa cell lysate and can be used at less than or equal to 5 µg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          Purity: Greater than 90%, as determined by SDS-PAGE.

          Aggregation: Less than 10%, as determined by HPLC.

          Filtration: 0.2 µm post-manufacturing filtered.

          Target Information

          The directed movement of membrane-bounded organelles is an important process for various cellular functions including membrane transport, secretion, and axonal transport. Cytoplasmic dynein, a ubiquitous minus end-directed microtubule-based motor protein, is believed to be responsible for the retrograde transport of membranous organelles from the synapse to the cell body in fast axonal transport. The cytoplasmic dynein complex is composed of two ~530 kDa heavy chains, and intermediate chains of ~74 kDa and ~53-59 kDa.
