Fructosamine-3-kinase related protein (FN3KRP) is located on chromosome 17q25.3, approximately 8 kb upstream of the FN3K gene. It shares 65% amino acid identity with FN3K and possesses an identical genomic organization, implying functional similarities; FN3KRP is suggested to play a role in enzymatic deglycation. Unlike FN3K, FN3KRP is more ancient, present in diverse organisms such as cyanobacteria, echinoderms, plants, and cold-blooded vertebrates. Studies have discovered a significant association between rs1046896 in FN3KRP and longevity, where the longevity allele C is linked to increased FN3KRP expression in whole blood. FN3KRP is hypothesized to phosphorylate glucitolamines, products of non-enzymatic glycation, indicating its involvement in crucial metabolic processes. The regulation of both FN3K and FN3KRP is notable for TATA-less and CAAT-less promoters, featuring homologous CpG islands and Sp1 binding sites. Meanwhile, both genes are widely expressed across human tissues, but FN3K demonstrates significantly higher levels in tissues prone to glycation-related complications.
