Gamma-Glutamyl hydrolase (GGH) is a homodimeric protein that catalyzes the cleavage of the gamma-glutamyl chain of folylpoly-gamma-glutamyl substrates and is a central enzyme in folyl and antifolyl poly-gamma-glutamate metabolism. The activities of the baculovirus-expressed enzyme and the enzyme from HL-60 cells were similar on methotrexate polyglutamates. GGH is expressed in a variety of human tumors and correlates with poor prognosis.
