          Flow cytometry analysis of CD49d in THP-1 cells (green) compared to an isotype control (blue). Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/mL, fixed with 2% paraformaldehyde and washed with PBS. Cells were blocked with a 2% solution of BSA-PBS for 30 min at room temperature and incubated with a CD49d monoclonal antibody (Product # MA49D7) at a dilution of 0.5 µg/test for 60 min at room temperature.           MA49D7 targets CD49d in FACS, IHC, and Neu applications and shows reactivity with Rat and Human samples.



          The MA49D7 immunogen is rat 150 kD alpha4 chain of VLA-4 (CD49d).



          MA49D7 detects CD49d which has a predicted molecular weight of approximately 111 kDa.



          This product has been tested for endotoxins by limulus amoebocyte lysate (LAL) assay and contains an endotoxin concentration of less than or equal to 10 endotoxin units per milligram (EU/mg).

          Target Information

          The integrin alpha 4 (also known as CD49d and ITGA4) belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha and beta chains. Alpha4 chain associates with either beta1 or beta7 chain. It has been demonstrated that the putative ligand-binding sites of both integrin alpha4beta1 and alpha4beta7 is located on the alpha4 chain. These ligands included Madcam, VCAM, and fibronectin. Madcam is known as the principal ligand for integrin alpha4beta7. Recently it was also demonstrated that HIV-1 envelope can mimic Madcam by binding to and signaling though integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
