          Product Specific Information

          Description: The II/41 monoclonal antibody reacts with the mu heavy chain of mouse IgM. It does not react with other classes of mouse immunoglobulin including IgD, IgG or IgA. IgM is expressed intracellularly, during early stages of B lymphopoiesis, and then on the surface of more mature B cells in the bone marrow and peripheral B cells. Fluorochrome conjugated II/41 can be used as a detection secondary for mouse IgM.

          Applications Reported: This II/41 antibody has been reported for use in flow cytometric analysis.

          Applications Tested: This II/41 antibody has been tested by flow cytometric analysis of mouse bone marrow cells. This can be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

          PE-eFluor® 610 can be excited with laser lines from 488-561 nm and emits at 607 nm. We recommend using a 610/20 band pass filter (equivalent to PE-Texas Red®). Please make sure that your instrument is capable of detecting this fluorochome.

          Light sensitivity: This tandem dye is sensitive to photo-induced oxidation. Please protect this vial and stained samples from light.

          Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) (100 µL of cell sample + 100 µL of IC Fixation Buffer) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.

          Excitation: 488-561 nm; Emission: 607 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser.

          Filtration: 0.2 µm post-manufacturing filtered.

          Target Information

          Mouse IgM is an immunoglobulin M antibody that is produced by B cells in response to an antigen. It is the first antibody produced during an immune response and is the largest immunoglobulin in the serum. Mouse IgM is composed of five heavy chains and five light chains, and it is involved in complement activation and agglutination of antigens. It plays a crucial role in the innate immune response and is important for neutralizing pathogens before the adaptive immune response is fully activated. Mouse IgM is widely used in research and diagnostic applications, including ELISA, Western blotting, and immunohistochemistry.
