          Product Specific Information

Description: The monoclonal antibody 26mNOD2 reacts with mouse NOD2. NOD2, also referred to as caspase recruitment domain-containing protein 15 (CARD 15), is a cytosolic protein of approximately 115kDa. NOD2 contains a central NOD (nucleotide oligomerization domain) that is flanked by a C-terminal leucine-rich repeat (LRR) domain and an N-terminal CARD domain. The LRR confers recognition of microbial components such as muramyl dipeptide and the CARD domain confers the ability to interact with RIP2 via homotypic interaction with the RIP2 CARD. This interaction leads to downstream activation of NF-kappaB.

The antibody 26mNOD2 has been shown to recognize protein from transgenic and transfected cells. Refer to references for expression in endogenous cells.

Applications Reported: This 26mNOD2 antibody has been reported for use in immunoblotting (WB).

Applications Tested: This 26mNOD2 antibody has been tested by immunoblotting of lysates of 293T cells transfected with mouse NOD2. This antibody has been validated on transfected cell lines and cells from NOD2-transgenic mice.

Purity: Greater than 90%, as determined by SDS-PAGE.

Aggregation: Less than 10%, as determined by HPLC.

          Filtration: 0.2 µm post-manufacturing filtered.

          Target Information

NOD2 is a member of the apoptosis regulating protein family that includes caspase recruitment-domains, as well as Apaf-1 and NOD1. It contains two N-terminal CARDs, a nucleotide binding domain (NBD), and multiple C-terminal leucine-rich repeats (LRRs). NOD2 is expressed in monocytes (whereas NOD1 is expressed in multiple tissues). NOD2 plays a role in regulating NF-kappaB, acts as an intracellular receptor for bacterial lipopolysaccharides, and contributes to inflammatory bowel disease (IBD) and Crohn's disease.
