          PKLR Antibody (PA5-34734) in WB

          Western blot was performed using Anti-PKLR Polyclonal Antibody (Product # PA5-34734) and bands between 58-62 kDa band corresponding to PKLR was observed in K-562 and Hep G2, but not in U-2 OS, U-937 and Raji which are reported to be low expressing cell lines. Whole cell lysate (30 µg lysate) of K-562 (Lane 1), Hep G2 (Lane 2), U-2 OS (Lane 3), U-937 (Lane 4) and Raji (Lane 5) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX).           Target Information

          The protein encoded by this gene is a pyruvate kinase that catalyzes the transphosphorylation of phohsphoenolpyruvate into pyruvate and ATP, which is the rate-limiting step of glycolysis. Defects in this enzyme, due to gene mutations or genetic variations, are the common cause of chronic hereditary nonspherocytic hemolytic anemia (CNSHA or HNSHA). Multiple transcript variants encoding different isoforms have been found for this gene.
