          Phospho-Asap1 (Tyr782) Antibody (600-401-910) in ICC/IF

Immunofluorescent microscopy using Rocklands Affinity Purified anti-ASAP1 pY782 antibody shows detection of phosphorylated ASAP1 present in mouse NIH3T3 cells transfected with activated Src. Specific staining is not present when antibody is pre-incubated with the immunizing peptide prior to reaction with cells. Personal Communication. Product Specific Information

Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

This affinity-purified antibody is directed against the phosphorylated form of mouse ASAP1 protein at the pY782 residue. Reactivity occurs against mouse ASAP1 pY782 protein and the antibody is specific for the phosphorylated form of the protein. Reactivity with non-phosphorylated mouse ASAP1 is minimal by ELISA. A BLAST analysis was used to suggest cross reactivity with ASAP1 proteins from human, chicken, bovine, dog, rat and chimpanzee based on 100% homology with the immunizing sequence. Reactivity against homologues from other sources is not known.

          Target Information

ASAP1, also known as Development and differentiation enhancing factor 1, is a protein with two ANK repeats, an Arf-GAP domain, a PH domain and a SH3 domain. ASAP1 is a homodimer known to interact with SRC and CRK and its activity is stimulated by phosphatidylinositol 4,5-biphosphate (PIP2). It possesses phosphatidylinositol 4,5-biphosphate-dependent GTPase-activating protein activity for ARF1 (ADP ribosylation factor 1) and ARF5 and a lesser activity towards ARF6. It may coordinate membrane trafficking with cell growth or actin cytoskeleton remodeling by binding to both SRC and PIP2 and may also function as a signal transduction protein involved in the differentiation of fibroblasts into adipocytes and possibly other cell types. It is widely expressed in most of the tissues. ASAP1 is a protooncogene involved in invasive breast cancer and in other types of invasive and malignant tumors, such as uveal melanomas.
