Semaphorin 5B (SEMA5B) is a gene belonging to the Semaphorin family, which is known for its role as guidance cues in various biological processes. The gene is located on chromosome 3q21.3 and encodes for the transmembrane protein Semaphorin 5B, which participates significantly in neural development and cancer processes. SEMA5B serves as a guidance cue in neural tissues, impacting axonal growth and branching by acting through receptors like PlexinA1. It is proteolytically processed from its transmembrane form to a soluble form, capable of collapsing growth cones in neurons, thereby regulating neuronal pathfinding. Studies have revealed that SEMA5B impacts sensory afferent projections through interactions with cell adhesion molecules such as TAG-1. Moreover, SEMA5B has been implicated in cancer, particularly showing upregulated expression in kidney renal clear cell carcinoma (KIRC) at both mRNA and protein levels. Its expression correlates with various clinical parameters, including tumor stage and patient demographics. Its dynamic expression pattern in the spinal cord suggests a role in the modulation of sensory axon growth, further emphasizing its importance in developmental neurobiology and pathology.
