AA Sequence of recombinant protein: AEKPSPCQCS RLSPHNRTNC GFPGITSDQC FDNGCCFDSS VTGVPWCFHP LPKQESDQCV MEVSDRRNCG YPGISPEECA SRKCCFSNFI FEVPWCFFPK SVEDCHY.
Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human TFF-2. Anti-Human TFF-2-specific antibody was purified by affinity chromatography and then biotinylated.
Sandwich ELISA: To detect Human TFF-2 by sandwich ELISA (using 100 µL/well) a concentration of 0.25-1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti- Human TFF-2 (500-P312) as a capture antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human TFF-2.
Western Blot: To detect Human TFF-2 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. When used in conjunction with compatible development reagents the detection limit for Recombinant Human TFF-2 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
500-P312BT-1MG will be provided as 2 x 500 µg
Trefoil peptides are protease resistant molecules secreted throughout the gut that play a role in mucosal healing and protection of the gastrointestinal epithelia. These peptides contain three intrachain disulfide bonds, forming the trefoil motif, or P-domain. SP (spasmolytic polypeptide), also designated Trefoil factor 2 (TFF2) precursor, is a trefoil protein that functions to inhibit gastrointestinal motility and gastric acid secretion. SP may also act as a structural component of the gastric mucus, possibly by stabilizing glycoproteins in the mucus gel through interactions with carbohydrate side chains. A downregulation of SP expression is associated with primary gastric cancer, and a progressive loss of this protein is likely to be involved in the early stage of the multi-step gastric carcinogenesis pathway.
