UCP4 Antibody (PA5-100668) in ICC/IF
Immunocytochemistry analysis of UCP4 in HepG2 cells. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25°C. Samples were incubated with UCP4 polyclonal antibody (Product # PA5-100668) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies. The nuclear counter stain is DAPI (blue).

Target Information
UCP4 is a 323 amino acid mitochondrial uncoupling protein belonging to the large mitochondrial substrate carrier family with three Solcar repeats. UCP4 is a mitochondrial transport carrier that facilitates the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. It reduces the mitochondrial membrane potential in mammalian cells and is involved in thermoregulatory heat production. UCP4 creates proton leaks across the inner mitochondrial membrane, thus uncoupling oxidative phosphorylation from ATP synthesis resulting in dissipation of energy in the form of heat. UCP4 is usually found in adult and fetal brain, with low levels in spinal chord, corpus callosum and substantia nigra.
