ZNF554, or Zinc Finger Protein 554, is a member of the Kruppel-associated box domain-zinc finger proteins (KRAB-ZNF) subfamily, residing on chromosome 19p13. This region is characterized by a concentration of heterochromatin proteins, which indicates tightly regulated gene expression. ZNF554 is implicated in various cellular processes including transcription regulation and apoptosis. Dysregulation of ZNF554 has been linked to several diseases; notably, its decreased expression is associated with poor prognosis in gliomas, specifically grades III and IV astrocytomas, where it affects the PI3K-Akt signaling pathway. It also plays a role in placental development, with dysfunction contributing to conditions such as fetal growth restriction (FGR). Additionally, ZNF554 transcription is influenced by factors like ZEB1, ZEB2, and hypoxic conditions, with neighboring hypoxia-response elements showing hypermethylation in preeclampsia. This gene's expression can significantly alter the transcriptional landscape, with overexpression leading to the differential regulation of hundreds of genes, underscoring its importance in cellular regulation.
