Immunofluorescent analysis of ZO-2/TJP2 Antibody (3E8D9) was done on 90% confluent log phase CACO2 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. ZO-2 Antibody — Figure 2. ZO-2 down-regulates cyclin D1 transcription. (A) Cyclin D1 promoter activity was tested under conditions of high ZO-2 levels. MDCK cells were transiently cotransfected with 1 mug of pXP2-CD1 reporter plasmid and the indicated amounts of pGW1-HA-ZO-2 (ZO-2) expression vector or with the empty vector (pGW1). pXP2-CD1/pGW1 is taken as control. (B) To test the specificity of the ZO-2 effect, MDCK sparse monolayers were cotransfected with different reporter plasmids (pRSV-beta-Gal, pCAT-Sox, or pXP2-CD1) and ZO-2 or pCB6 ZO-1myc (ZO-1) as indicated in the graph. Control reference activity was taken for each reporter as the activity under the influence of pGW1. (C) Cells were transiently transfected with 10 nM siRNA designed to target ZO-2 mRNA or a NC siRNA not homologous to anything in the vertebrate transcriptosome. Forty-eight hours after initial transfection cells were retransfected with 1 mug of pXP2-CD1 reporter plasmid. Harvesting was performed after 72 h (total time). Graphs show relative luciferase activity taken as a percentage of control (only pXP2-CD1) activity. (D and E) In vivo transcription of cyclin D1 tested in response to differential expression of ZO-2. Total RNA was extracted from monolayers transiently transfected with full-length ZO-2 (pGW1-HA-ZO-2), the empty vector (pGW1; D), or transfected with siRNA for ZO-2 or NC (E). Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) ubiquitously expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. CD45 is absent on non-hematopoietic cell lines, normal and malignant, non-hematopoietic tissues. CD45 glycoprotein is crucial in lymphocyte development and antigen signaling, serving as an important regulator of Src-family kinases. CD45 protein exists as multiple isoforms as a result of alternative splicing, differ in their extracellular domains but share identical transmembrane and cytoplasmic domains. CD45RA is an isoform of the CD45 complex and has restricted expression between different subtypes of lymphoid cells. CD45 isoforms differ in their ability to translocate into the glycosphingolipid-enriched membrane domains and their expression depends on cell type and physiological state of the cell. CD45 has been shown to be an essential regulator of T- and B-cell antigen receptor signaling and suppresses JAK kinases to regulate cytokine receptor signaling. CD45 is also important in promoting cell survival by modulating integrin-mediated signal transduction pathway, DNA fragmentation during apoptosis and inhibition or upregulation of various immunological functions.
