Invitrogen Platinum Hot Start PCR Master Mix, available in colorless and green formats, offers Platinum Taq DNA polymerase in an optimized PCR buffer with magnesium and dNTPs for convenient PCR setup. It retains all the features of Platinum Taq DNA Polymerase: specificity, robustness, and reliability.

Platinum Green Hot Start PCR Master Mix additionally contains a density reagent and two tracking dyes for direct loading of PCR products on gels.

Platinum Hot Start PCR Master Mix provides a ready-to-use mixture of DNA polymerase, salts, magnesium, and dNTPs, thus reducing the number of pipetting steps during PCR reaction setup. It is supplied with a separate vial of Platinum GC Enhancer, formulated for specific amplification and improved yields of GC-rich targets.

Platinum Taq DNA Polymerase is a recombinant Taq DNA polymerase complexed with proprietary antibodies that inhibit enzyme activity at room temperature, reducing nonspecific target amplification at lower temperatures. The antibodies dissociate during the initial PCR denaturation step, and DNA polymerase regains its full activity. Just as with Taq DNA Polymerase, Platinum Taq DNA Polymerase has a non-template-dependent terminal transferase activity that adds a 3' deoxyadenosine to product ends and has a 5' to 3' exonuclease activity.

Do you have any recommendations for troubleshooting the amplification of GC-rich or problematic targets with Platinum Hot Start PCR Master Mix (2X) (Cat. Nos. 13000012, 13000013, 13000014)?

We have the following recommendations:
Increase the extension time from 1 min/kb to 1.5 min/kb.
Vary total PCR cycles from 20-40; 35 cycles is typical.
Try denaturing at 95 degrees C for 45 seconds.
GC-rich or problematic targets work better with MgSO4 instead of MgCl2.
For GC-rich templates, test higher annealing temperatures starting with the temperature that is equal to your primer Tm and in increments of 2 degrees C for up to six different temperatures.

Note: Addition of Platinum GC Enhancer solution enhances the amplification of GC-rich and problematic sequences.

What is the final concentration of MgCl2 in the Platinum Hot Start PCR Master Mix (2X) (Cat. Nos. 13000012, 13000013, 13000014)?

The final concentration is 1.5 mM MgCl2.

When should the GC enhancer be used that comes with the Platinum Green Hot Start PCR 2X Master Mix (2X)?

We recommend using the GC enhancer for targets with >65% GC sequences. We recommend that you have a final concentration of 20% of the GC enhancer in the reaction mix. There is enough GC enhancer included in the kit to add it to every PCR reaction.

Is the green dye that comes with the Platinum Taq Hot Start Green DNA Polymerase and Platinum Green Hot Start PCR Master Mix (2X) compatible with downstream applications?

Yes the green dye is compatible with downstream applications such as fluorescent automatic DNA sequencing, ligation, and restriction digestion.
