MagMAX™ DNA Multi-Sample Kits are designed for rapid purification of high-quality genomic DNA from a variety of sample types including blood and blood cards, tissue, mouse tails, buccal swabs, buffy coats, and cultured cells. These purification kits use MagMAX™ magnetic bead-based nucleic acid isolation technology to produce high yields of purified DNA, free from inhibitors that may affect downstream reactions such as genotyping assays, CE and next-generation sequencing, and high resolution melting analysis. The 50 prep kit is designed for manual microfuge tube processing.

The demands for DNA yield and purity have become more stringent with the development of new analytical methods and technologies, such as high-throughput genotyping, and next-generation DNA sequencing platforms. MagMAX™ DNA Multi-Sample Kits meet the challenge by delivering highly pure DNA that is free of nucleases, proteins, and other inhibitors of downstream enzymatic reactions.
As shown in Table 1, DNA with high yield and purity can be obtained from a variety of materials using the MagMAX™ DNA Multi-Sample Kits. This is in contrast to filter membrane–based methods that often require different kit configurations for various input materials. For applications requiring maximal yields, for small batch sizes, or for DNA archival purposes MagMAX™ DNA Multi-Sample Kit (tube format, manual processing) may be desirable because larger sample inputs are accommodated. DNA yields suitable for most routine applications may be obtained using the MagMAX™-96 DNA Multi-Sample Kits (96-well plate format, compatible with automated handling) (Table 1).

The MagMAX™ DNA Multi-Sample Kit also delivers high yields of DNA from lipid or glycogen rich samples, such as brain and liver tissues, respectively. Figure 1 shows more than twice as much DNA was purified from tissues with the MagMAX™ DNA Multi-Sample Kit than with the leading competitor’s filter membrane–based method.

Most enzymatic molecular applications are prone to inhibition by analytes carried over from the sample matrix (e.g., heme, heparin, salts, protein) or the purification method itself (phenol, salts, alcohol). Figure 2 illustrates the effect of DNA purity on sensitive molecular applications, such as quantitative PCR (qPCR). DNA was extracted from blood using the MagMAX™-96 DNA Multi-Sample Kit, or a competitor’s filter membrane–based kit and subjected to real-time PCR analysis. The cycle threshold CT values obtained with the DNA purified using the MagMAX™ DNA Multi-Sample Kit were lower than those obtained with the filter membrane kit–purified DNA, indicating the MagMAX™ kit yields higher-purity DNA.

MagMAX™ DNA Multi-Sample Kits provide purified DNA that is suitable for a variety of Applied Biosystems application workflows (Figure 3), including SNP genotyping and DNA sequencing analysis (see Figure 4). What is the difference between Proteinase K for MagMAX DNA Multi-Sample Ultra Kit (Cat. No A25561) and Proteinase K for MagMAX 96 DNA Multi-Sample Kit (Cat. No. 4489110)?

The difference between Proteinase K for MagMAX‐96 DNA Multi‐Sample Kit and Proteinase K for MagMAX DNA Multi‐Sample Ultra Kit is the brand. The former is from Invitrogen legacy and the latter is from Applied Biosystems legacy. Both products have an identical concentration of 100 mg/mL and can be used interchangeably.

I am using the MagMAX DNA Multi-Sample kit on my MagMAX Express-96 instrument and notice that I start with 100 microliters elution volume and end up with only 80 microliters. How can I prevent loss of the elution volume?

The MagMAX beads will cause a loss of ~2-3 µL of solution when they are pulled from the solution, but this is a very small amount. Larger volume loss, such as 20µL, can be due to several factors:
• Low lab humidity combined with a heated elution can cause this amount of liquid to evaporate. In this case, we recommend increasing the starting elution volume. Since the nucleic acid is not evaporating this should not affect your overall yield.
• Issues with alignment of the instrument components could also result in loss of volume. If the head goes down too deep during mixing, it could pull droplets up along the side. This would reduce elution volume and you would also be more likely to see a reduction in yield. A similar issue would result if the turntable alignment was off on either side.

Can I edit the manufactured KingFisher Flex protocols?

No, manufactured protocols are locked for editing, but they can be renamed using the ‘save as' function and edited as a new version.

Can I use magnetic beads from other vendors with the KingFisher Flex instrument?

For nucleic acid extraction, we recommend using our MagMAX kits and MagJET kits. Other magnetic beads should work well as long as they are 0.5 to 10 µm in size. However, you will have to write your own KingFisher protocols using the Thermo Scientific BindIt software and validate/optimize the protocol on your own.
