The new Ion Xpress™ RNA-Seq Barcode 1-16 Kit provides a set of 16 unique barcode primers for sequencing RNA on the Ion Torrent PGM™ semiconductor sequencer. When used in combination with the Ion Total RNA-Seq Kit v2 (Cat. No. 4475936), the Ion Xpress™ RNA-Seq Barcode 1-16 Kit enables users to pool up to 16 fragment libraries prior to template preparation and then conduct multiplexed sequencing analysis, simplifying the Ion semiconductor sequencing workflow for small and whole transcriptome RNA.

Sequence- and Flow-Optimized for Increased Performance and Efficiency

Barcode primers contain sequences that are optimized to provide equal representation of all barcodes in a pool, require a minimum number of flows to interrogate, and necessitate only 20 bases of barcode and adaptor sequencing, which results in more economical multiplexed sequencing runs.

Built-in error correction significantly minimizes the possibility of incorrect sample assignment.

Why does the Ion Total RNA-Seq Kit v2 (Cat. No. 4475936) specifically require the Ion Xpress RNA-Seq Barcodes 1-16 Kit (Cat. No. 4475485)? Why can't I use the DNA barcodes provided in the Ion Xpress Barcode Adaptors 1-96 Kit (Cat. No. 4474517)?

The Ion Total RNA-Seq Kit v2 workflow uses a different method for adding the barcode sequences to the library than the standard Ion DNA libraries. For standard DNA library construction, the barcoded adapters are directly ligated to the DNA inserts. For Ion Total RNA-Seq libraries, the barcode adapter sequence is added during the final library amplification.

How many samples can I multiplex on a single chip for sequencing on the Ion PGM or Proton System?

The number of samples that can be multiplexed in a single sequencing run depends on the capacity of the chip, the size of the library, and the required coverage.
