This PrimeFlow™ RNA Assay is an in situ hybridization assay that combines the power of branched-DNA technology with the single-cell resolution of flow cytometry, enabling the simultaneous detection of up to four RNA targets in combination with immunophenotyping for cell surface and intracellular proteins using fluorochrome-conjugated antibodies. Branched-DNA technology uses pairs of target-specific probes to amplify the detection of a RNA transcript, achieving between 8,000- to 16,000-fold signal amplification from a desired RNA transcript. PrimeFlow RNA Assay can detect mRNA, long non-coding RNA, viral RNA, and telomere DNA. The detection of microRNA is also possible and is optimal when using the PrimeFlow™ microRNA Pretreatment Buffer (cat. 88-18006) and protocol. PrimeFlow RNA Assay is compatible with human and mouse cells. Other mammalian and non-mammalian cells may be compatible

This PrimeFlow RNA Assay contains all the reagents needed to conduct the assay. Positive control probes and target probes for genes of interest are sold separately.

Do you supply positive control probes as part of the PrimeFlow RNA Assay, Cat. No. 88-18005-204, 88-18005-210?

Positive control probe sets are sold separately. Please refer to Appendix 5 of the PrimeFlow RNA Assay User Manual for recommendations and catalog numbers for recommended positive control genes. However, the appropriate positive control gene for your specific model system should be determined empirically.

What is the sensitivity for microRNA detection with the PrimeFlow RNA Assay kit with the miRNA Pretreatment Buffrer (Cat. No 88-18006)?

The sensitivity is estimated to be 40 copies per cell with Type 1, Alexa Fluor 647 Probe Sets. The actual sensitivity may vary depending on the specific target and Probe Type.

Is the microRNA Pretreatment buffer (Cat. No. 88-18006) required for microRNA detection in the PrimeFlow RNA Assay?

The Pretreatment Buffer is not absolutely required for microRNA detection. However, it can enhance detection for most of the microRNA targets tested.

How does the data look for microRNA staining if we use the standard PrimeFlow RNA Assay protocol (Cat. No. 88-18009 or 88-18001)?

In some cases, there is a decrease in the signal for a given microRNA target, in other cases there is no difference. The extent of the decrease and whether or not it occurs varies depending on the specific microRNA target being analyzed.
