Gibco Dynamis liquid medium is specifically designed to offer the highest batch and fed-batch culture performance and yield with recombinant CHO cells in a chemically defined environment. Gibco Dynamis  medium has also been shown to support robust growth of HEK293 cells for adenovirus production. The chemically defined, protein-free, animal origin component-free formulation provides the power to achieve high titers, start process development faster and streamline or simplify transfer to manufacturing scale.

Power to achieve higher titers
Dynamis Medium is fortified to help maximize recombinant CHO cell growth and protein expression in suspension batch culture, offering 170% greater production than select competitive media with simple glucose supplementation by day 14. Cultures grown in Dynamis Medium extend to even higher titers demonstrating good cell viability out to day 21. This medium can also provide up to 60% greater fed-batch growth and production over glucose-only fed-batch culture for selected cell lines and applications when used with EfficientFeed C+ Supplement.

Power to start process development faster
As a fortified medium, Dynamis provides the power to get to a high cell density over 10 million cells per mL faster than competitive products and sustains that cell density longer by just maintaining glucose levels. The viable cell growth achieved make it a good choice for scale-up if you have started with CD FortiCHO medium. When paired with EfficientFeed C+ Supplement, it provides a fast and efficient way to develop a process.

When to Use Dynamis Medium
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S cell line
• The cell line you are using is HEK293 for adenovirus production
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

When to Consider Another Gibco Medium

Consider CD OptiCHO Medium when:
• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky,' i.e. hard to grow or adapt

Consider CD CHO Medium when:
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

Consider CD DG44 Medium when:
• You use parental DG44 or DXB11 CHO cells (dhfr-)

Does Dynamis Medium contain a surfactant? Do I need to add extra surfactant for a suspension CHO culture in a shake flask?

While the full formulation for Dynamis Medium is proprietary information that cannot be disclosed, we can confirm that the medium contains a surfactant and that no additional surfactant would need to be added to the medium to support your suspension CHO cells in shake flask culture.

How well do cells created or frozen in CD FortiCHO medium thaw into Dynamis medium for growth and production?

CD FortiCHO medium has proven to be an excellent single-stream medium for CHO production clone development and banking. In our experience, CHO clones created and selected in CD FortiCHO medium can be put directly into Dynamis medium without any adaptation. In fact, our work suggests that those cells will produce as well or better in Dynamis medium and are just as stable in Dynamis medium if used for process development and scale-up.

If I've done preclinical work in CD FortiCHO medium and want to transition to Dynamis medium, can that be done without repeating all the earlier work?

We have done some studies to show comparability of results between the two media, and that information may be filed with agencies in support of any work your Regulatory Team feels is necessary to establish transitioning from the one medium to the other. It is possible that characterization of your expressed protein after you see no other titer changes will be viewed no differently than a CMC update or minor modification to feeding profile during technical transfer to manufacturing. Of course our Regulatory Team would be happy to have a discussion with your team if it makes this transition any simpler for you.

What feeding supplementation is recommended with Dynamis medium?

When culturing cells without complete feeds for fed-batch production, we recommend that you do not to allow glucose levels to fall below 2 g/L in culture by supplementing with glucose as needed (see product insert for more details). We have found that EfficientFeed A+, B+, and/or C+ supplements work well with Dynamis medium to maximize titers. We recommend trying EfficientFeed C+ 1X and 2X first, and if specific productivity drops off late in culture try the addition of FunctionMAX Titer Enhancer. No supplements should be added before reaching the day when half-peak cell density is reached for cultures not fed with complete feeding supplements like the EfficientFeed supplements.
