The Human Dopaminergic Neuron Immunocytochemistry Kit enables optimal image-based analysis of three key markers of neural differentiation: OTX2, FoxA2, and tyrosine hydroxylase (TH). This high-performance immunocytochemistry (ICC) kit includes a complete set of primary and secondary antibodies, a nuclear DNA stain, and premade buffers for an optimized staining experiment.

Confirm expression of key cellular markers
The antibodies included in the kit have been carefully selected and validated for high performance in the ICC analysis of floor plate progenitors and dopaminergic neurons. OTX2 and FoxA2 are critical nuclear markers while TH is an important cytoplasmic marker.

The antibody combinations in the kit have been designed to allow for the specific and simultaneous assessment of two markers at a time (e.g., OTX2 and FoxA2, and FoxA2 and TH) along with nuclear DNA staining to help save time and precious sample.

The streamlined method recommended for this kit eliminates unnecessary wash steps found in standard ICC protocols, helping to save time and decrease the likelihood of cell loss during the staining protocol. The only washes required are after incubation with primary and secondary antibodies.

Frequently asked questions (FAQs)

The Human Dopaminergic Neuron Immunocytochemistry Kit provides the NucBlue Fixed Cell Stain. If I don't have the correct filter set for this stain, can I use another nucleic acid stain?

If you use Alexa Fluor 488/Alexa Fluor 555 dye-conjugated secondary antibodies and you have a Cy5/Alexa Fluor 647 filter set, you may use a far-red nucleic acid stain, such as NucRed Dead 647 ReadyProbes Reagent (Cat. No. R37113) or SYTOX Deep Red Nucleic Acid Stain (Cat. No. S11380). Do not use these far-red nucleic acid stains if using an Alexa Fluor 594 dye-conjugated secondary antibody as there is too much spectral overlap of Alexa Fluor 594 into the Cy5/Alexa Fluor 647 range.

We do not recommend using the Human Dopaminergic Neuron Immunocytochemistry Kit for flow cytometry analysis. The kit and protocol were specifically developed for imaging analysis (microscopy/high-content screening (HCS)).

Do you offer the individual components of the Human Dopaminergic Neuron Immunocytochemistry Kit as standalone products?

Only the NucBlue Fixed Cell ReadyProbes Reagent may be purchased separately as Cat. No. R37606. We do offer the secondary antibodies separately, but not in the same proprietary concentration as provided in the kit. We also offer formaldehyde, DPBS, BSA, and saponin as separate catalog numbers, but not in the volumes or as formulated in the kit. The primary antibodies are not available as separate stock items; all non-stock items in the kit may be available as a custom order. Please contact your Account Representative.
