Quantitative & consistent recovery
Quantitative and consistent recovery of DNA from challenging matrices—including samples that have high protein concentration, low pH, or high salt—often presents a technical challenge. The PrepSEQ nucleic acid extraction kits are now adapted for higher-throughput workflows (up to 96 extractions, or 24 samples in triplicate) when used in combination with the KingFisher Flex 96 Deep-well Head Magnetic Particle Processor. The special formulation of magnetic particles provided in the PrepSEQ kits help ensure efficient and specific recovery of DNA from challenging samples.

High quality & throughput isolation
Most automated liquid handling systems move reagents into and out of a single well to perform the different steps of nucleic acid isolation. In contrast, the KingFisher Flex 96 Deep-well Head Magnetic Particle Processor utilizes permanent rods to collect the magnetic beads from solution and releases the beads into a well containing reagents for the next step of isolation. The effectiveness of bead collection and transfer leads to superior washing, elution efficiency, and processing. The KingFisher Flex 96 Deep-well Head Magnetic Particle Processor can process up to 24 samples in triplicate, compared to 3 samples in triplicate using the manual method.

Compatible with PrepSEQ sample preparation kits
The KingFisher Flex 96 Deep-well Head Magnetic Particle Processor has been optimized to work with both the PrepSEQ Residual DNA Sample Preparation Kit and the PrepSEQ 1-2-3 Nucleic Acid Extraction Kit assays. The instrument comes pre-installed with PrepSEQ optimized protocols and does not require any further optimization after installation and validation.

My Pharma KingFisher Flex run was stopped due to an error message. Can I start the protocol from where it stopped?

No, once a protocol was stopped, it will restart from the beginning. The extraction procedure can either be finished manually by using a magnetic stand, or a modified version of the original Pharma KingFisher Flex script would have to be created that starts off at the point where the run was stopped.

Is it always necessary to use the transport locks on the Pharma KingFisher Flex instrument?

The transport locks are only necessary when relocating the instrument.

Are the volumes of reagents in each well critical for a successful Pharma KingFisher Flex run?

We strongly recommend that you maintain the specified volumes within the defined limits to avoid spillover and to help ensure the best performance at the most efficient level.

What if the magnetic particles are attached to the tip combs after my Pharma KingFisher Flex run?

This happens sometimes but it will not affect the yield because the sample has been released from the particles.

What if the magnetic particles remain in the sample well during a Pharma KingFisher Flex run?

If the starting material is too viscous, the magnetic rods will not be able to collect the particles. Dilute the sample and check that the sample is properly homogenized/lysed. Adding low amounts of detergent will improve the collection of magnetic particles as well. Quickly centrifuge the plate to sediment the particles to the bottom of the plate.
