Gibco Primary Rat Cortex Neurons, Sprague Dawley, are isolated and cryopreserved from Sprague Dawley embryonic day-18 rats. A flexible, ready-to-use, and quality alternative to fresh neurons, each vial contains one million cells per mL. These neurons consistently yield post-thaw viability in the range of 50–80%, have high neuronal purity, and are validated for functionality.

High viability: post-thaw viability of 50% or greater
High purity: greater than 70% neuronal phenotype
Functionally validated: tested for spontaneous synchronous calcium flux and synaptic release activity

Frequently asked questions (FAQs)

What is the difference between Primary Rat Cortex Neurons, Sprague Dawley and Primary Rat Cortex Neurons?

These neurons differ in the rat strain from which they were isolated: Sprague Dawley vs. Fisher 344. These neurons don't expand anymore. They just grow neurites. The manual (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/MAN0001574-PrimaryRatCortex-HippocampusNeurons-UG.pdf) has instructions for culturing the cells. You can use Neurobasal Plus/B-27 Plus/GlutaMAX to culture them, or just Neurobasal/B-27/GlutaMAX. However, the former combination provides much superior performance than the latter and can maintain neurons longer.

I'm currently culturing neurons. Which media system do you recommend using in conjunction with the Poly-D-Lysine matrix?

We recommend using the B-27 Plus Neuronal Culture System when culturing neurons on Poly-D-Lysine. The B-27 Plus Neuronal Culture System is a complete media system that includes the B-27 Plus Supplement and the Neurobasal Plus Medium.

What cell types will Poly-D-Lysine support as an attachment matrix?

The Poly-D-Lysine matrix has successfully supported the attachment of primary rodent neurons, iPSC-derived neurons and PC12 cells.

Can primary rat neurons be cryopreserved again after they are cultured?

Primary rat neurons cannot be cryopreserved again because they will not revive.

Can primary rat neurons proliferate?

No, primary rat neurons do not proliferate.
