Ponceaus S Staining Solution is a ready-to-use membrane stain for evaluating the transfer efficiency of a western blot. This stain is compatible with nitrocellulose and PVDF membranes. Ponceau S staining is reversible and can be removed with a short incubation in 0.1% NaOH.
Ponceaus S Staining Solution is a ready-to-use membrane stain for evaluating the transfer efficiency of a western blot. This stain is compatible with nitrocellulose and PVDF membranes. Ponceau S staining is reversible and can be removed with a short incubation in 0.1% NaOH.

Features include:
• A nondestructive, reversible method to stain and detect proteins on nitrocellulose and PVDF membranes
• Minimal nonspecific interactions with the membrane surface; provides a reliable method for visualization of protein transfer to a membrane
• Simple and quick staining protocol that results in reddish-pink bands that are easily photographed
• Reversed easily in less than five minutes
• Subsequent western blot detection unaffected as the stain does not alter the membrane-bound protein

Frequently asked questions (FAQs)

What is the best way to image Ponceau staining with an iBright Imaging System?

The recommended imaging method depends on the iBright imager model being used. For iBright FL1500 and CL1500 Imaging Systems, we recommend imaging Ponceau-stained samples with the Ponceau application that can be found in Universal mode. For the iBright CL750 Imaging System, we recommend imaging Ponceau-stained samples in Protein gel mode.

We always recommend imaging with the orange iBright System Contrast Tray (Cat. No. A56597), and if needed, adjust the focus or zoom to get bands as sharp as possible. Better quality pictures are obtained when the membrane is wet.

Can I use the No-Stain Protein Labeling Reagent with an orange or red dye (i.e., will the signal appear in channel 2, 3, 4 or 5, not just channel 1 in the iBright imager)?

Channels 3, 4 and 5 on the iBright imager can be used for multiplexing without interference while using the No-Stain Protein Labeling Reagent. Channels 1 and 2 on the iBright imager will show interference. Fluorophores for multiplexing that are compatible with the No-Stain Protein Labeling Reagent are listed here, under the “Secondary Abs” tab (https://www.thermofisher.com/us/en/home/life-science/protein-biology/protein-assays-analysis/western-blotting/detect-proteins-western-blot/total-protein-normalization.html).
