          1. Immobilized Recombinant Human CD86/B7-2 Protein (Fc Tag) at 2 µg/mL (100 µL/well) can bind Human CTLA-4 His-tag Recombinant Protein, the EC50 is 2.6-8 ng/mL.
          2. Measured by its ability to inhibit IL-2 secretion by stimulated Jurkat human acute T cell leukemia cells. The ED50 for this effect is 0.05-0.3 µg/mL when stimulated with 1 µg/mL Recombinant Human B7-1/CD80.
          3.Captured Ipilimumab on proA Chip can bind Human CTLA-4 His-tag Recombinant Protein with an affinity constant of 20.03 nM as determined in a SPR assay (Biacore T200)(Routinely tested).
          4.Loaded Ipilimumab on ProA Biosensor, can bind Human CTLA-4 His-tag Recombinant Protein with an affinity constant of 5.15 nM as determined in BLI assay (Sartorius Octet Red384) (Routinely tested).

          Reconstitution: Dissolve the protein in sterile double distilled water to a concentration of 0.2 mg/mL or lower. It is recommended that the protein be aliquoted and be used as soon as possible. Store aliquots under sterile conditions at -20°C. Avoid repeated freeze-thaw cycles.

          Target Information

          CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed on mainly CD4+ T cells. It binds the same ligands as CD28 (CD80 and CD86 on B cells and dendritic cells), but with higher affinity than CD28. However, in contrast to CD28 which enhances cell function when bound at the same time as the T cell receptor, CTLA4 inhbits the T cell and prevents it from functioning. Intracellular CTLA4 is also found in regulatory T cells and may be important to their function.
