Increasing Analytical Depth via Native LC-MS Biopharmaceutical Characterization

Native mass spectrometry has emerged as a powerful tool to understand protein assemblies and post-translational modifications (PTMs) in structural biology. The coupling of liquid phase separations such as size exclusion chromatography (SEC-MS), or cation or anion exchange chromatography (IEX-MS) with native MS, facilitates biopharmaceutical characterization for the analysis of aggregates and size variants of monoclonal antibodies (mAbs).

The ability to perform native LC-MS has been driven by advances in high-resolution mass spectrometry (HRMS) and the associated bioinformatics used for post-acquisition spectral processing. In this webinar, Drs. Füssl and Carillo will present an evaluation of the performance of Thermo Scientific hybrid quadrupole-Orbitrap MS instruments, namely the Thermo Scientific Q Exactive Plus mass spectrometer and the Thermo Scientific Orbitrap Exploris 240 mass spectrometer for the characterization of mAbs and other biopharmaceuticals, using native LC-MS.

Florian Füssl received his Master of Science degree and his Ph.D. from the University of Salzburg, Austria. In 2016, he started as postdoctoral researcher in the Characterization and Comparability Laboratory of Dr. Jonathan Bones at NIBRT in Dublin, Ireland. His main focus is the development of novel methods for the characterization of biopharmaceuticals and proteins in general using liquid chromatography-mass spectrometry (LC-MS) techniques with special emphasis on intact protein analysis.

Dr. Sara Carillo is the Applications Development Team Leader in the Characterization and Comparability Laboratory in NIBRT, Ireland. Sara completed her Ph.D. in Chemical Science at the University of Naples, Italy, in 2013 working on bacterial glyco-conjugates structural characterization. In 2015, Sara joined Dr. Jonathan Bones’ research group in NIBRT, working on CHO cell glycome and biotherapeutics structural characterization.
